Jagged1 regulates endometrial receptivity in both humans and mice

چکیده

The human endometrium undergoes cycle-dependent changes and is only receptive to an implanting blastocyst within a narrow window of 2-4 days in the mid-secretory phase. Such functional require delicate interplay between diversity factors including cytokines signaling pathways. Notch pathway members are expressed endometrium. We have previously demonstrated that ligand Jagged1 (JAG1) localizes endometrial luminal epithelium (LE) abnormally reduced infertile women during receptivity. However, consequences JAG1 production on receptivity implantation unknown. This study aimed determine role regulating humans mice. Knockdown both primary epithelial cells Ishikawa significantly their adhesive capacity HTR8/SVneo (trophoblast cell line) spheroids. confirmed cells, interacted with Receptor 3 (NOTCH3) knockdown expression downstream target HEY1 classical markers. Jag1 mouse LE impaired implantation. identified ten genes (related tight junction, infertility, adhesion) were differentially by Further analysis junction family species revealed altered components Together, our data regulated mice likely via different mechanisms.

Redefining Endometrial Receptivity

However, the notion of receptivity as a static state in which the tissue is poised, frozen in time, awaiting the arrival of an adequately developed embryo, is naïve. An integrative definition of receptivity must embrace communication between maternal and embryonic tissues. At receptivity, the endometrium has reached a point at which negotiation can begin that may or may not lead to successful i...

P-224: Mechanisms That Reduce Endometrial Receptivity in Endometriosis

Background: Several studies indicate clear relationship between infertility and endometriosis. However, the etiology of this association is uncertain. Endometrial dysfunction is a possible cause of infertility in women with endometriosis, which could explain the high rates of implantation failure and miscarriages in these women. Delay in endometrial maturation and intrauterine biochemical abnor...

Endometrial Receptivity: Cell Biological Aspects

The cell biological basis of endometrial receptivity and trophoblast invasiveness, the two preconditions for implantation ("implantation windowtl), still remains enigmatic. Recently, however, the application of modern concepts of cell biology and developmental biology concentrating on celladhesion and cell polarity seems to open promising new views of these phenomena. At implantation initiation...
